Hi, I'll review this article. Will have comments up in a few days. Sasata (talk) 21:02, 30 September 2013 (UTC)[reply]

Comments

  • lead: suggested links – activated fatty acid, mitochondrial membrane, beta-oxidation, expression, microarray
Done. Adabow (talk) 22:40, 7 October 2013 (UTC)[reply]
  • the enzyme infobox is in an unfortunate location, squeezing the accompanying text, which makes the biochemical reaction harder to read; I suggest moving this info box to the right hand side underneath the main infobox
  • could we have a little more background about the significance of the reaction; it's not until later we learn that the enzyme is involved in "lysine degradation", but I think this should be made explicit earlier
  • "Similar to many other 2OG oxygenases, the activity of gamma-butyrobetaine dioxygenase can be stimulated by reducing agents such as ascorbate and glutathione.[7][8][9][10]" why are four sources needed for this statement?
  • "The catalytic activity of gamma-butyrobetaine dioxygenase can be stimulated with different metal ions, especially potassium ions." this is sourced to a paper from 1988; in the intervening time, has there been any research into the nature of this stimulatory effect?
  • "Both the apo (PDB id: 3N6W)" for a general encyclopaedia article, one should assume the reader will not know biochemical jargon, so PDB should be spelled out and linked on 1st occurrence
  • "demonstrating an induced fit mechanism may contribute to the catalytic activity of gamma-butyrobetaine dioxygenase." source?
  • "promiscuis" spell correctly and link; also link substrate selectivity
Promiscuous done; no WP article for substrate selectivity. Adabow (talk) 22:40, 7 October 2013 (UTC)[reply]
  • "natural catalytic product L-carnitine and D-carnitine" products
  • check article for repeated links
  • "Some studies suggested that mildronate may also be beneficial for the treatment of neurological disorder,[22][23] diabetes,[24] and seizures and alcohol intoxication.[25]" this is a sweeping generalization; is this MEDRS compliant?
  • "To date, at least five clinical trial reports were published in peer-reviewed journals documenting the efficacy and safety of mildronate on the treatments of angina, stroke and chronic heart failure.[26][27][28][29][30]" I don't think these citations are necessary or appropriate for an article on the enzyme; is there a secondary source which summarizes these reports?
  • link binding pocket, non-competitive
Done. Adabow (talk) 22:40, 7 October 2013 (UTC)[reply]
  • where's the history? Who first reported the enzyme? The article only talks about the human version of the enzyme. Has it been reported in other mammals? How close are the sequences?
  • the article states the enzyme structure has been determined, but there is no discussion about its structure. Is is monomeric or multimeric? Any conformational changes with binding? Is it a soluble enzyme or located in the membrane? What is its MW? Isoelectric point?
  • the last sentence of the lead mentions that "BBOX1 has recently been identified as a potential cancer gene on the basis of a large-scale microarray data analysis." but this is not discussed anywhere else (a violation of WP:LEAD); please give details
Not much movement on this GAN, so I'm closing it. Hopefully these comments will be useful to other editors who wish to improve the article. Sasata (talk) 17:32, 14 October 2013 (UTC)[reply]
