Overall

  • I think the article is well-written and well-organized. Although I'm not an expert on RNA pol II, I it seems that you have a lot of good information.
  • There are some places where I think scientific terms could use more explanation. I'll note these in my section-by-section suggestions.

Discovery of RNApol II

  • Although scientifically-literate people know what the nucleolus and nucleoplasm are, I don't know if laymen would.
  • I changed a sentence in this section ("In 1969, science experimentalists...") to make it more clear.
  • It might be helpful to briefly explain ion exchange chromatography.

Subunits

  • It might help to explain the implications to the core enzyme's inability to recognize specific promoters, e.g. whether or not transcription can occur.
  • It would be helpful to include a diagram of RNApol II that shows where the subunits are. I know there are different RNApol II's, so maybe use human or yeast?

Assembly

  • A diagram of the assembly process would be very helpful.

Kinetics

  • The first paragraph here seems tangential. The article as about RNApol II, not enzymes in general.
  • Perhaps instead you could say what the average Km and Kcat values are for RNApol II enzymes.
  • What are alpha-amanitin and amatoxins?

Holoenzyme

  • I think you should cite the last sentence of paragraph one, as well as paragraph 2.

Control by chromatin structure

  • The structure of this section is a little confusing. I think it would benefit from a topic sentence that says a general statement about the relationship between chromatin structure and RNApol II.
  • There are no citations in this section.

Protein complexes involved

  • At the top, is that a list? If so, I would use bullet points.
  • Images of the CTD domain would fit well here.
